Absorbance can support a concentration result when the analytical method, sample preparation and calibration relationship are defined. The instrument does not identify every substance in a sample simply by measuring its colour.

In water and wastewater laboratories, many spectrophotometric tests use reagents to form a coloured reaction product. Selecting an instrument therefore involves more than choosing a wavelength range. The method, sample cell, reagent system, expected concentration and measurement location all matter. Two relevant instruments listed on the Artin Azma website are the portable Hach DR1900 and the laboratory Hach DR6000. Their exact optical arrangement varies between instruments. A general explanation of these components should not be mistaken for a service drawing of a particular model.

The source produces light. The wavelength selection system directs a selected portion of that light towards the sample. After passing through the sample cell, the light reaches the detector. The instrument compares the measurement with its reference condition and calculates transmittance or absorbance. A concentration calculation then requires the calibration relationship associated with the selected method.

Transmittance is defined as T = I / I₀, where I is transmitted intensity and I₀ is reference intensity. Absorbance is A = −log₁₀(T). When transmittance is expressed as a percentage, the equivalent expression is A = −log₁₀(%T / 100). Absorbance is dimensionless, although instrument displays commonly identify it with Abs.

Under conditions where the Beer–Lambert relationship is valid, absorbance is proportional to concentration and optical path length. Real samples may depart from those conditions. Chemical interferences, background colour, scattering and unsuitable calibration ranges can prevent a simple conversion. A stable absorbance reading is therefore only one part of a valid analytical result.

Hach DR1900 and DR6000: the main differences

Feature

Hach DR1900

Hach DR6000

Instrument type

Portable VIS spectrophotometer

Laboratory UV–VIS spectrophotometer

Wavelength range

340–800 nm

190–1100 nm

Light source

Xenon flash lamp

Deuterium for UV and tungsten-halogen for the visible region

Selection context

Measurements at the sampling location using compatible methods

Laboratory methods requiring a broader wavelength range

Table 1. Document who approved the procedure and how later changes will be assessed.

Sample preparation and measurement timing

Sampling is part of the measurement process. An unsuitable container, delayed testing or incorrect preservation may alter the sample before it reaches the instrument. Correct instrument operation cannot reconstruct the original sample. Obtain container requirements, storage conditions and permitted holding times from the method for the individual parameter; there is no single preservation instruction suitable for all tests.

Retain the sampling location, collection time and any preparation details with the sample record. Where sampling and analysis are performed by different people, make the handover information explicit. A result with an unclear sample identity remains difficult to interpret even if the optical measurement was performed correctly.

For colour-forming reactions, track reagent addition and reading times. When a method specifies a colour development or stability interval, an earlier or later reading is not automatically equivalent. Use appropriate volumetric equipment for small additions. Incomplete mixing may cause uneven response, while excessive shaking may introduce bubbles. Apply the mixing instructions of the actual test.

Dilution and the final reported concentration

Use an authorised diluent and suitable volumetric equipment. Dilution may reduce an interference, but it does not guarantee its elimination. It can also move an analyte towards the lower end of the method’s working range. Confirm that the diluted solution gives a result within the applicable interval and respect any method-specific restrictions on dilution.

Hypothetical example: bringing 10 mL of sample to a final volume of 50 mL gives a dilution factor of 5. If the diluted solution reads 2 mg/L and the instrument has not already applied that factor, the original sample concentration is 10 mg/L. Applying the factor both in the instrument and again manually creates an incorrect result.

Make the point of application explicit in the laboratory procedure. Record both the dilution preparation and the calculation so another analyst can reconstruct the result. Zeroing establishes a reference. Instrument verification examines characteristics such as wavelength accuracy or photometric response. Method calibration defines the relationship between response and analyte concentration under the specified analytical conditions.

Use suitable reference materials or filters with relevant certificates and acceptance limits. A calibration procedure written for a UNICO instrument should not be transferred to a DR1900 or DR6000 without confirmation. Internal adjustment is not a substitute for investigating a failed performance check. Establish the cause and the authorised corrective action before changing settings.

The appropriate checks depend on the intended use and the laboratory quality programme. A control standard with an assigned value can help assess performance, but acceptance depends on the method and laboratory quality programme. One fixed percentage is not automatically appropriate for every analyte and concentration. Check standard suitability, validity and storage conditions before interpreting a failure.

Duplicate samples provide information about result dispersion. A matrix spike can help investigate recovery where the method supports it. Passing a standard prepared in clean water does not establish the absence of interference in an actual wastewater sample. These checks answer different questions and should be selected for the problem being investigated.

Retain failed controls and the corrective action record. Repeatedly reading a control until an acceptable number appears is not an investigation. Hach’s guidance on checking colorimetric accuracy discusses the distinct contributions of instrument performance, reagents and sample conditions.
